The aim of this study was to analyze the toxicities and autostimulatory activities of a phytohemagglutinin (PHA)-containing liposome preparation in vitro, and to determine its effects on CD4+ and CD8+ T cells, T cell proliferation, cytotoxic activity, and natural killer (NK) cell activity. The PHA-containing liposome was prepared by the direct lyophilization method and was encapsulated in a polyethylene glycol stearate (stearic acid) aqueous solution. The toxicities of the liposome were evaluated by indirect fluorescent antibody staining (FA/IFAS) and the results indicated that the cytotoxic activities of PHA-containing liposomes, at doses of 5-10 microg/ml, were reduced to 4.1% and 3.6% respectively, and the MHC-unrestricted and MHC-restricted T cell activities of peripheral blood mononuclear cells (PBMCs), at doses of 5-10 microg/ml, were reduced to 4.7% and 4.0%, respectively, while the NK cell activity of PBMCs, at doses of 5-10 microg/ml, were reduced to 4.5%. The toxicities of the liposome were observed at an early stage of the in vitro culture. Thus, it is very important to use PHA-containing liposomes at a small dose in clinical applications.
